Challenges in Traditional Exosome Quantification Methods
Early methods for exosome characterization primarily focused on their physical properties.

(1)Exosome Morphology Identification: Transmission Electron Microscopy (TEM)→Requires expensive equipment and trained personnel for operation.

(2)Exosome Size and Concentration Determination: Nanoparticle Tracking Analysis (NTA) →Non-specific screening.

It has become clear that relying solely on physical characterization is insufficient to meet all quality control requirements for exosomes.
As exosome quality control gains increasing importance, the identification of exosome surface proteins has become a critical need

Exosome surfaces are rich in

(1)Transmembrane proteins involved in transport, such as CD9, CD63, and CD81.

(2)Heat shock proteins (HSP60, HSP70, HSP90), and cell-specific proteins.

While CD9, CD63, and CD81 are commonly used exosome surface biomarkers, recent literature indicates that CD81 is not exclusively presented on exosomes.
